Which characters could appear in S.H.I.E.L.D.? Eric Goldman at IGN speculates on which Marvel characters might make an appearance. And of course, the inevitable "which actors should appear in it?" articles have starting popping up all over the place. Here's one from the Huffington Post.

Give it another day and there will be plenty of "Why Joss should write S.H.I.E.L.D. exactly the way I want it" pop-ed pieces.

I think SHIELD should consist of a bunch of characters Joss makes up, with references to the known Marvel universe.
I'd love to see Bradley Whitford and Richard Jenkins team-up again for the show.

I want a lot of superhero involvement. What's a marvel universe show that doesn't have superhero involvement?
At the moment I'm really impressed with Marvel and their ideas of expanding the verse. They seem to have a solid master plan. Having a S.H.I.E.L.D. TV show at the core, they can actively promote every new film with that, if it's well done. Even a little forshadowing would get the fans excited. I just hope they get to tell their own tales as well.
But if Marvel would be a Marvel "big bad", we'd be doomed.
Can't argue with Summer, Enver, and Tricia Helfer. Please no, on some of the others.
I trust that the right people will get parts big and small, since Joss will work with casting people who understand his and his crew's way of working. That said...I definitely think Summer and Enver could do with a bit more Joss-powered mojo to get things moving (especially with Summer, since Knights of Badassdom needs to be released and getting some big buzz from her being in a major TV series would help :D).
